Recombinant human Tumor Necrosis Factor-a His produced in
E. coli is a single, non-glycosylated, polypeptide chain containing 157 amino acids fragment (77-233) and having a molecular mass of 18.3 kDa with an amino-terminal hexahistidine tag. The TNF-Alpha His is purified by standard chromatographic techniques.
Synonyms: TNF-Alpha, Tumor necrosis factor ligand superfamily member 2, TNF-a, Cachectin, DIF, TNFA, TNFSF2.
Formulation: TNF-a His is supplied in 1xPBS.
Physical Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.
Purity: Greater than 97% as determined by SDS-PAGE.
Source:Escherichia coli. Uniprot ACC number: P01375
Transportation method: Shipped at room temperature.
Stability: Lyophilized TNF-a although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution TNF-a should be stored at +4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.
Biological Activity: EC50 0.200-0.120 ng/ml measured in a metabolic inhibitor actinomycin D assay using L-cells (ATCC CCL-1).
Solubility: It is recommended to reconstitute the lyophilized TNF-α in sterile 18MΩ-cm H
2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions.
