The Immortalized Mouse Prostate Epithelial Cells (iMPEC, clone7) is derived by co-expression of adenovirus E1A and dominant-negative p53 (p53DD). In addition to its non-tumorigenicity, this cell line maintains its primary phenotype and expresses tissue-specific markers such as the androgen receptor and the homeobox protein Nkx3.1. This cell line is a useful model for studying prostate growth regulation. Immortalization Method: Serial passaging and electroporation transfection with linearized DNA of E1A and p53DDBioSafety Level: IISpecies: MouseSource Organ: Prostate glandsGrowth Properties: AdherentMorphology: CuboidalPopulation Doubling: 23 - 33 hoursSeeding Density: 25,000 - 30,000 cells/cm²Markers: Androgen receptor, Nkx3.1, β-catenin, CK8, CK18 and vimentinDonor Age: N/ADonor Gender: N/ADonor Ethnicity: N/APropagation: The base medium for this cell line is Prigrow III medium available from abm (TM003). To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to final concentration of 10% and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%. Carbon dioxide (CO2): 5%, Temperature: +37.0°C. * abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.Quality Control: 1. Western blot was used to confirm the presence of E1A and p53DD gene in the immortalized cell line. 2. Western blot was used to check tissue-specific markers.Shipping Condition: Dry IceStorage Condition: liquid nitrogen or -180°C
