The Immortalized Mouse Cardiomyogenic Progenitor Cells are hygromycin-resistant, non-tumorigenic reversibly immortalized cells that have cardiomyocyte-like morphology and have maintained rhythmic beating in culture. It expresses late stem cell markers (Oct3/4, Nanog, Sox2), c-kit, early cardiomyogenic markers (ISL-1, Hand2, Sca-1), and cardiomyocyte-specific markers (α-actin and α-myosin heavy chain). The immortalization process may be reversed by infection with Cre-recombinase expressing adenoviruses (abm Cat. No. 000018A) to revert immortal cells back into primary cells resulting in increasing differentiation potential. In the presence of retinoic acid, the cells undergo cardiomyogenesis where late cardiomyogenic markers (Actc1 and ANF) will be present. Dexamethasone may be used as a differentiation reagent, to express late markers ANF, cTnT, and α-MyHC. These cells are recommended for studies relating to cardiomyogenic progenitors in basic and/or translational research.Immortalization Method: Serial passaging and transduction with retrovirus SSR#69 expressing SV40 large T antigen flanked with Cre/loxP sitesBioSafety Level: IISpecies: MouseSource Organ: HeartGrowth Properties: AdherentMorphology: PolygonalPopulation Doubling: N/ASeeding Density: Thaw entire contents into an appropriate T25 flask as specified in the Propagation instructions.Markers: N/ADonor Age: N/ADonor Gender: N/ADonor Ethnicity: N/APropagation: The base medium for this cell line is Prigrow III medium available at abm (TM003). To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to final concentration of 10% and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%.
