Immortalized Human Mammary Epithelial Cells (HMEC 2.6)-SV40 is an immortalized breast epithelial cell line which are not tumorigenic and do not show no anchorage-independent growth. There is a normal distribution of cells in G1, S, and G2/M phase and they express normal breast epithelial markers ( E-cadherin, CK7/18, and CK5/14). The cells retain primary characteristics and are valuable for research relating to mammary epithelial biology.Immortalization Method: Serial passaging and transduction with recombinant lentiviruses carrying SV40 Large T antigenBioSafety Level: IISpecies: HumanSource Organ: BreastGrowth Properties: AdherentMorphology: PolygonalPopulation Doubling: N/ASeeding Density: N/AMarkers: Puromycin resistanceDonor Age: N/ADonor Gender: N/ADonor Ethnicity: N/APropagation: The base medium for this cell line is PriGrow IV medium available at abm, Cat. No. TM004. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 1% or 35 µg/ml bovine pituitary extract (Hammond), 12.5 ng EGF (Z100135), 50 µM fresh ascorbic acid, 2 nM estradiol, 1 µg/ml insulin, 2.8 µM hydrocortisone, 0.1 mM ethanolamine, 0.1 mM L-glutamine (G275), 15 nM sodium selenite, 1 ng/ml cholera toxin, and Penicillin/Streptomycin (G255) to a final concentration of 1%. For selection use G418 at 100 µg/ml and Zeocin at 20 µg/ml. Adjust media pH to 7.4.
Carbon dioxide (CO2): 5%, Temperature: +37.0°C.* abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.Quality Control: Real Time PCR was used to quantify SV40 gene expression in immortalized cell line.Shipping Condition: Dry IceStorage Condition: liquid nitrogen or -180°C
