Immortalized Human Mammary Epithelial Cells (HMEC 2.6) is an immortalized breast epithelial cell line which are not tumorigenic and do not show no anchorage-independent growth. There is a normal distribution of cells in G1, S, and G2/M phase and they express normal breast epithelial markers ( E-cadherin, CK7/18, and CK5/14). The cells retain primary characteristics and are valuable for research relating to mammary epithelial biology.Immortalization Method: Serial passaging and transduction with LXSN-based retroviruses carrying hTERT and a mutant CDK4(R24C)BioSafety Level: IISpecies: HumanSource Organ: Mammary TissueGrowth Properties: AdherentMorphology: PolygonalPopulation Doubling: Mean doubling time of around 72 hours.Seeding Density: 10,000 - 15,000 cells/cm²Markers: E-cadherin, EGFR, MIB1, CD10, CK5/14, P63, CK7/18, G418 resistanceDonor Age: N/ADonor Gender: FemaleDonor Ethnicity: N/APropagation: The base medium for this cell line is Prigrow IV (TM004). To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 1% or 35 µg/ml bovine pituitary extract (Hammond), 12.5 ng EGF (Z100135), 50 µM fresh ascorbic acid, 2 nM estradiol, 1 µg/ml insulin, 2.8 µM hydrocortisone, 0.1 mM ethanolamine, 0.1 mM L-glutamine (G275), 15 nM sodium selenite, and Penicillin/Streptomycin Solution (G255) to a final concentration of 1%. For selection use G418 at 100 µg/ml and Zeocin at 20 µg/ml. Adjust media pH to 7.4. Carbon dioxide (CO2): 5%, Temperature: +37.0°C.* abm does not recommend to use heat-inactivated FBS for cell culture unless specified otherwise.Quality Control: 1) Cell markers were probed via immunohistochemical analysis; 2) Soft agar assay preformed to determine anchorage-independence.
