Immortalized Human Adult Brain Astrocytes (AST-1) is immortalized via the introduction of only the cytoplasmic content (non-DNA) of tumor cells into primary human brain astrocytes, thereby resulting in permanent growth activation. One key aspect of such immortalization method is its resemblance of the immortalized cells to its primary counterparts (i.e. grows in monolayer). AST-1 cells can be used for exploring the diversity of astrocytes functions, including regulation of neurogenesis and control of synaptic functions.

Propagation:Use of PriCoatTM T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.
The base medium for this cell line is Prigrow IV medium available at abm, Cat. No. TM004. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (TM999)* to a final concentration of 5%, Penicillin/Streptomycin Solution (G255) to a final concentration of 1%, Recombinant Human FGF2 (Z101455) to a final concentration of 10 ng/ml, Recombinant Human GH1 (Z100265) to a final concentration of 10 ng/ml, and Recombinant Human HGF (Z102775) to a final concentration of 10 ng/ml.Change media every 2-3 days.Carbon dioxide (CO2): 5%, Temperature: +37.0°C.* Do not use heat-inactivated FBS for cell culture unless specified otherwise.
